Excimer Based Molecular Beacons
Molecular beacons (MBs) are fluorescent nucleic acid probes with a stem-loop structure. In our approach, we have developed an excimer-controlled MB with non-nucleosidic chromophores in the stem. Excimer fluorescence is inhibited by formation of a donor-acceptor complex between alkynylpyrene and perylenediimide units results in an excellent reduction of background signal, which allows the sensitive detection of target DNA and RNA even in the presence of significant excess of the probe. In combination with bright excimer fluorescence target detection is rendered possible at low nanomolar concentrations.
